该研究以库页红景天细胞水提物(Rhodiola sachalinensis A.Bor.cell extract,HCE)为材料,评估其对铜绿假单胞菌和猪霍乱沙门氏菌的抑菌作用及对呼吸代谢的影响。结果表明:HCE对2种菌的抑制效果显著。在铜绿假单胞菌中,HCE在与碘乙酸(iodoacetic acid,IA)、丙二酸(malonic acid,MA)、十二水合磷酸三钠(trisodium phosphate dodecahydrate,TSP)3种典型呼吸抑制剂共同作用的呼吸叠加率测量实验中,HCE与MA的呼吸叠加率最低;在猪霍乱沙门氏菌中,HCE与TSP的呼吸叠加率最低。对铜绿假单胞菌与猪霍乱沙门氏菌对应的糖酵解途径(embden-meyerhof-Parnas,EMP)与磷酸戊糖途径(hexose-monophosphate-pathway,HMP)中关键酶活性的检测表明,HCE能够通过调控EMP途径中磷酸果糖激酶(phosphofructokinase,PFK)和丙酮酸激酶(pyruvate kinase,PK)的表达抑制铜绿假单胞菌的呼吸代谢过程,并通过调节HMP途径中葡萄糖-6-磷酸脱氢酶(glucose-6-phosphate dehydrogenase,G6PD)的表达抑制猪霍乱沙门氏菌的呼吸代谢过程,进而降低铜绿假单胞菌和猪霍乱沙门氏菌的活性。该研究探明了HCE通过呼吸代谢对铜绿假单胞菌和猪霍乱沙门氏菌活性的影响,为HCE的应用及开发奠定了基础。
Abstract:This study evaluated the antibacterial effects and respiratory metabolic influence of Rhodiola sachalinensis A.Bor.cell extract(HCE) on Pseudomonas aeruginosa and Salmonella choleraesuis.The results showed that HCE was effective in inhibiting the two bacteria, in the assays for respiratory stacking rates, three representative respiratory inhibitors were used: iodoacetic acid(IA), malonic acid(MA) and trisodium phosphate dodecahydrate(TSP), the lowest respiratory stacking rate was observed in Pseudomonas aeruginosa for HCE and MA; and the lowest respiratory stacking rate was observed in Salmonella choleraesuis for HCE and TSP.The activity of key enzymes in embden-meyerhof-parnas(EMP) and hexose-monophosphate-pathway(HMP) showed that HCE could inhibit the respiratory metabolic process of Pseudomonas aeruginosa by regulating the expression of phosphofructokinase and pyruvate kinase in EMP pathway, and inhibit the respiratory metabolic process of Salmonella choleraesuis by regulating the expression of glucose-6-phosphate dehydrogenase in HMP pathway, and then reduce the activity of Salmonella choleraesuis process, which in turn reduced the activity of Pseudomonas aeruginosa and Salmonella choleraesuis.
