3种药物对弓形虫感染小鼠的治疗效果研究

摘要:

为确定青蒿素、黄芩苷、桦褐孔菌多糖对弓形虫感染BALB/c小鼠的治疗效果,该实验将80只BALB/c小鼠随机分为8组,每组10只。模型组和模型给药组小鼠每只腹腔注射弓形虫RH株速殖子103个。正常给药组连续灌胃给药14d,模型给药组在攻虫24h后连续灌胃给药14d。实验期间每日称量小鼠体重,观察饮食量及临床症状,给药14d后脱颈处死各组小鼠,采用酶联免疫吸附实验(enzyme-linked immuno sorbent assay,ELISA)检测血清中免疫球蛋白亚类和细胞因子水平,HE染色镜检小肠组织病理变化。结果表明:青蒿素、黄芩苷、桦褐孔菌多糖在标准浓度下对正常小鼠饮食量、临床症状及生长发育等均无显著影响;3种药物对IL-4、IFN-γ、Ig G1、Ig G2a水平以及小肠器官无显著影响,对消化系统无毒性作用;3种药物对弓形虫感染模型小鼠作用差异显著,青蒿素、黄芩苷可显著提升弓形虫感染小鼠血清Ig G2a含量。该研究证实青蒿素、黄芩苷及桦褐孔菌多糖在浓度分别为200、100、200mg/kg时,对健康小鼠机体安全性良好;青蒿素、黄芩苷抑制弓形虫效果较好,桦褐孔菌多糖抑制弓形虫效果不显著。该研究为青蒿素、黄芩苷及桦褐孔菌多糖制剂的临床合理用药提供了毒理学依据,并为青蒿素、黄芩苷及桦褐孔菌多糖对弓形虫的抑制作用提供了参考。

Abstract:

To determine the therapeutic effects of artemisinin,baicalein,and Inonotus obliquus polysaccharides on Toxoplasma infected BALB/c mice,80 BALB/c mice were randomly divided into 8 groups,with 10 mice in each group.Mice in the model group and the model drug administration group were intraperitoneally injected with 103 tachyzoites of Toxoplasma gondii RH strain.The normal drug administration group was continuous intragastric administration for 14 days,and the model drug administration group was continuous intragastric administration for 14 days after 24 hours of infection.During the experiment,the body weight of the mice was measured daily,and the food intake and clinical symptoms were observed.After 14 days of administration,the mice in each group were sacrificed by cervical dislocation.The levels of immunoglobulin subclasses and cytokines in the serum were detected by enzyme-linked immuno sorbent assay(ELISA),and the pathological changes of the small intestine tissue were examined by HE staining.The results showed that artemisinin,baicalein,and Inonotus obliquus polysaccharides exhibit no significant effects on the food intake,clinical symptoms,and growth and development of normal mice at the standard concentrations.The three drugs present no significant effects on the levels of IL-4,IFN-γ,IgG1,IgG2a,and the small intestine organs,and display no toxic effects on the digestive system.The three drugs demonstrate significant differences in their effects on Toxoplasma gondii-infected model mice.Artemisinin and baicalein could significantly increase the serum IgG2a content in Toxoplasma gondii-infected mice.This study confirmed that artemisinin,baicalein,and Inonotus obliquus polysaccharides were safe for healthy mice at concentrations of 200mg/kg,100 mg/kg,and 200 mg/kg,respectively.Artemisinin and baicalein had better inhibitory effects on Toxoplasma gondii,while Inonotus obliquus polysaccharides reveal no significant inhibitory effect.This study provides toxicological evidence for the rational clinical use of artemisinin,baicalein,and Inonotus obliquus polysaccharides preparations and provides a reference for the inhibitory effects of artemisinin,baicalein,and Inonotus obliquus polysaccharides on Toxoplasma gondii.
